腺样囊性癌 TrkC 差异表达的 PDX 模型的建立与 鉴定

目的:建立并鉴定腺样囊性癌(adenoid cystic carcinoma,ACC)关于原肌球蛋白受体激酶 C(trypomyosin-receptor kinase C,TrkC)差异表达的人源肿瘤异种移植模型(patient-derived tumor xenograft,PDX)。方法:收集北京同仁 医院头颈外科收治的腺样囊性癌患者的新鲜肿瘤组织,皮下接种至免疫缺陷鼠体内,并不断传代至新一批的小鼠体 内直至肿瘤稳定生长。分析患者 ACC 肿瘤组织在免疫缺陷动物体内自然生长状态下的组织病理特征。结果:本次 实验共收集 ACC 标本 10 例,其中 5 例成功建立 PDX 模型,3 例完成鉴定。建立的 PDX 模型病理形态和组织结构与 患者肿瘤组织保持一致;3 例 PDX 模型 TrkC 表达呈现不同的特点。结论:PDX 模型可以很好地保留患者的肿瘤异 质性,有助于深入研究 ACC 发病机理,为以 TrkC 为靶点的临床前药物筛选提供动物模型。

[Abstract] Objective:To establish and identify a patient-derived tumor xenograft (PDX)model for the differential expression of TrkC in adenoid cystic carcinoma (ACC).Methods:Fresh tumor tissue of patients with ACC treated in the department of otolaryngology in Beijing Tongren Hospital were collected,subcutaneously inoculated into immunodeficient mice, and continuously passed into a new batch of mice until the tumors stably grew.The histopathological characteristics of ACC tissue in natural state of immunodeficient animals were analyzed.Results:Ten ACC samples were collected in this experiment.PDX models were successfully established for 5 cases.Pathology,morphology and structure of established PDX models were consistent with those of patients’tumor tissue;TrkC expression in the 5 PDX models showed differ characteristics.Conclusion:The PDX model can well preserve the tumor heterogeneity,which is conducive to the in-depth study of the pathogenesis of ACC,and provides an animal model for preclinical screening of Trkc-targeted drugs.
